Do I need to let off of the nitrous when I am shifting gears with the air shifter, since the kill box is killing the motor for a split second?

No.
Although you will hear the tell-tale nitrous backfire as you hit the button to shift thru the gears...

I ran for a full season with no mproblems.never let off.If I turned my kill to min. time You could not hear
any backfire.Use a micro switch to activate it and leave it on the whole pass.

Also, I forgot to mention it is a wet system.

I run a wet system on my oil-cooled gix, and it doesn't pop at shift when the delay is short enough. The way mine works is the WOT switch activates the nitrous so there's no easy way to "let off" the nitrous during the kill-time. I dunno how you'd "let off" for that short of a time anyway... I do have a sensor on the transmission to prevent the nitrous from spraying in 1st gear though.
